Nitric oxide activates an enzyme called guanylate cyclase, which results in increased levels of cyclic guanosine monophosphate (cGMP). cGMP relaxes the smooth muscles in the blood vessels of the penis, allowing them to widen and increase blood flow to the erectile tissue, leading to an erection.
